نویسندگان

چکیده

This presentation will review basic and clinical concepts on pain mechanisms underlying opioid analgesia. It describe the structure, function, cellular signaling of receptors; endogenous exogenous receptor ligands; as well central peripheral sites actions. The also discuss novel opioid-based therapeutic strategies, developed from recently gained knowledge structures signaling, pathological situations, in vivo studies, aimed at reduction side effects such respiratory depression, constipation, addiction, sedation tolerance. Lastly, problems associated with use be addressed.

Modulation of Peripheral m-Opioid Analgesia by s1 Receptors

We evaluated the effects of s1-receptor inhibition on m-opioid– induced mechanical antinociception and constipation. s1-Knockout mice exhibited marked mechanical antinociception in response to several m-opioid analgesics (fentanyl, oxycodone, morphine, buprenorphine, and tramadol) at systemic (subcutaneous) doses that were inactive in wild-type mice and even unmasked the antinociceptive effects...

Opioid Receptors of the Central Amygdala and Morphine-Induced Antinociception

The amygdala is a forebrain region, which is known as a modulator of pain sensation. The amygdala, particularly the central nucleus, has high concentrations of enkephalins relative to dynorphins and has high concentrations of opioid receptors. We here studied the role of central nuclei of amygdala in morphine antinociception. Methods: In this study, we used 130 male Wistar rats (200- 250g). Bil...

Structure, function, and plasticity of GABA transporters

GABA transporters belong to a large family of neurotransmitter:sodium symporters. They are widely expressed throughout the brain, with different levels of expression in different brain regions. GABA transporters are present in neurons and in astrocytes and their activity is crucial to regulate the extracellular concentration of GABA under basal conditions and during ongoing synaptic events. Num...

Biochemistry of opioid (morphine) receptors: binding, structure and molecular modelling

Morphine is the most widely used compound among narcotic analgesics and remains the gold standard when the effects of other analgetic drugs are compared. The most characteristic effect of morphine is the modulation of pain perception resulting in an increase in the threshold of noxious stimuli. Antinociception induced by morphine is mediated via opioid receptors, namely the μ-type opioid recept...

Opioid Receptors Gene Polymorphism and Heroin Dependence in Iran

Introduction: Genes often have multiple polymorphisms that interact with each other and the environment in different individuals. Variability in the opioid receptors can influence opiate withdrawal and dependence. In humans, A118G Single Nucleotide Polymorphisms (SNP) on μ-Opioid Receptor (MOR), 36 G>T in κ-Opioid Receptor (KOR), and T921C in the δ-Opioid Receptor (DOR) have been...
